Competent E.Coli bacterial is most commonly used in DNA cloning and subcloning. The amount of ligated DNA is usually 10 ng per 100-200 ul of competent e.coli cells. Several vendors supply competent cells with different transformation efficiency (10^7-10^8 cfu/ug DNA).
For blue/white screening with cloning vector containing lacZ gene in MCS, use comination of IPTG and X-Gal.
