The mloar ration between insert and vector DNA is crutial for sucessful ligation. Most often we use insert:vector molar ration of 3:1. To calculate the amount of insert DNA (ng), use equation bellow.

(ng of vector x size of insert in kb/size of vector in kb) x (molar ratio of insert:vector)

DNA ligations are normally carried out in 10-20 ul volume with total 10-200ng DNA.
