Content: One step RT-PCR takes advantage of Thermus thermophilus (Tth) DNA polymerase that can reverse transcribe RNA into cDNA and amplify it subsequently. The error rate for Tth polymerase amplification is even higher than Taq therefore isn't ideal for PCR cloning. But with optimized condition to improve specificity, it can be used as PCR cloning method.
