Epitope Mapping Protocols offers more than thirty protocols for antibody production and mapping-all described in detail by leading experts and illustrated by proven examples from their own work. The methods range from those that use (or can use) whole, native antigens to those that use peptides and peptide libraries and those that require antigen expressed from recombinant DNA. A method for T-cell epitopes, instructions on using "natural" variants or mutants, and techniques for producing panels of antibodies for epitope mapping are also included.

Epitope Mapping covers all the major methods for the identification and definition of epitopes. The Pepscan assay is used to define B cell epitopes and makes use of synthetic peptides but can only be used if the amino acid sequence is known. It can be adapted for the delineation of both helper T cells and cytotoxic T cells. The identification of combined B and T cell epitopes can also be achieved using synthetic peptides. There are other methodologies for analysing for cytotoxic T cell epitopes such as the purification of antigens presented by MHC class I molecules and expression cloning. Site directed mutagenesis is also a powerful tool in epitope mapping and can be used to evaluate the role of single amino acids in immune complex formation. Protein footprinting makes use of monoclonal antibodies produced by hybridoma technology and relies on the fact that the epitope is protected from cleavage when bound as an antibody-antigen complex. It is only useful for small antigens. Other monoclonal antibody assays such as enzyme linked immunosorbent assay and haemaglutination and slot-blotting may also be used in epitope mapping. Random phage display libraries bring together the genetic and amino acid peptide sequence and can be screened with antibody and the resulting peptide DNA sequenced to confirm the amino acid sequence of a specific epitope. Investigation of carbohydrates can also be useful to epitope mapping as deglycosylation can lead to loss of antigenic activity. Epitopes are important to the pharmaceutical industry and wherever appropriate, pharmaceutical applications of the methods described are included. For each method there is a description of the technology, protocols, trouble-shooting, and advice on when to use the method. This book will therefore be invaluable to any researcher involved in epitope mapping.

Proteins interacting with diverse ligands - proteins, peptides or DNA - are the basic principles underlying many biological processes, such as antigen-antibody binding, signal transduction or receptor binding. The technique of oligopeptide synthesis on a cellulose membrane and the subsequent binding assays allow the investigation of protein interactions. A particular advantage of these peptide arrays (SPOT - technology) is the high number of oligopeptide probes that can be tested in parallel. Detailed protocols for peptide synthesis, and the analysis of protein-protein, protein-DNA interactions as well as epitope mapping are presented in this manual. It is ideally suited not only for basic research laboratories but also for diagnostic and therapeutic applications since many diseases are related to dysfunctions in protein recognition and binding.

Achievements and progress in genome mapping and the genomics of microbes supersede by far those for higher plants and animals, in part due to their enormous economic implication but also smaller genome size. In the post-genomic era, whole genome sequences of animal-associated microbes are providing clues to depicting the genetic basis of the complex host-pathogen relationships and the evolution of parasitism; and to improving methods of controlling pathogens. This volume focuses on a globally important group of intracellular prokaryotic pathogens which affect livestock animals. These include Brucella, Mycobacterium, Anaplasma and Ehrlichia, as well as the protozoan pathogens Cryptosporidium and Theileria, for which genome sequence data is available. Insights from comparative genomics of the microbes described provide clues to the adaptation involved in host-microbe interactions, as well as resources potentially useful for application in future research and product development.

This book describes basics of synthetic peptide technology and its usage in epitope mapping for disease causing viruses of human, animals and birds. It is handy for newcomers on how to analyse the genomic data of viruses and predict antigenic determinants also called as epitopes in structural proteins of viruses and how to fish out promising epitopes. The methods for using computer alogorithms like DNASTAR software and synthesizing peptides by simple methods were described lucidly.Preparation of multiple antigenic peptides, screening of peptides representing epitopes were stated very elaborately. Most importantly, the methods for producing antipeptide antibody in rabbits without using any carrier proteins which in turn will produce high titred sera was written in this book in simple language.Purification of sera by sepharose methods was also included in this book. This book is a compendium of all methods for Molecular Biology and Molecular Virology students, researchers and scientists. It is expected to fill the gap in synthetic peptide technology and its application for disease diagnosis and developing novel,safe,non-infectious immunogens as vaccines for controlling animal viral diseases.
